甲型流感病毒又称A型流行性感冒病毒（influenza A virus,IAV）属于正黏病毒科（Orthomyxoviridae），是单股负链分节段的RNA病毒。IAV基因组由8个节段构成，分别是聚合酶碱性蛋白2 (polymerase basic protein 2,PB2）、聚合酶碱性蛋白1 (polymerase basic protein 1,PB1）、聚合酶酸性蛋白(polymerase acid protein,PA)、血凝素(hemagglutinin,HA)、核蛋白（nucleoprotein,NP）、神经氨酸酶(neuraminidase,NA)、基质(matrix,M)蛋白和非结构蛋白（nonstructural protein,NS）节段。根据HA和NA的遗传性和抗原性的不同，可以将IAV分为18种HA亚型（H1-H18）和11种NA亚型（N1-N11）。
HA蛋白发挥其受体结合能力的前提是被宿主细胞蛋白酶（如跨膜丝氨酸蛋白酶TMPRSS2等）裂解为HA1和HA2两个亚基，而HA裂解位点氨基酸的不同，导致病毒的致病性也显著不同，同时HA蛋白的稳定性也能影响多种亚型流感病毒对家禽和哺乳动物的致病力。
