Belatacept 是一种重组融合蛋白，由细胞毒性 T 淋巴细胞相关抗原 4（CTLA-4）的胞外结构域与人 IgG1 的 Fc 段融合而成，通过哺乳动物细胞表达系统生产。它在 T 淋巴细胞表面有较高亲和结合位点，当机体免疫系统启动，尤其是在器官移植等免疫反应强烈的场景下，Belatacept 能够快速结合抗原呈递细胞表面的 CD80 和 CD86，阻断它们与 T 淋巴细胞表面 CD28 的相互作用，抑制 T 淋巴细胞的过度活化，避免免疫排斥反应过度发生。临床前研究和器官移植实验表明，Belatacept 能有效降低移植排斥反应，在肾移植模型中，它减少了免疫细胞对移植肾的攻击，提高了移植肾的存活率和功能，对器官移植免疫研究及抗排斥药物研发意义重大。
