T-cell surface antigen CD2（T 细胞表面抗原 CD2，也被称为 CD2、Erythrocyte receptor、SRBC、Rosette receptor、LFA-3 receptor、T-cell surface antigen T11/Leu-5、LFA-2 ），是 Research Grade Alefacept 的关键靶标。CD2 是一种广泛表达于 T 淋巴细胞表面的跨膜糖蛋白，在 T 细胞的活化、增殖以及免疫应答过程中发挥着至关重要的作用。在正常的免疫反应中，CD2 主要通过与抗原呈递细胞（APC）表面的配体 CD58（也称为 LFA3，lymphocyte function associated antigen 3）相互作用，为 T 细胞的活化提供重要的共刺激信号。这种相互作用能够增强 T 细胞对抗原的识别能力，促进 T 细胞分泌细胞因子，进而激活下游的免疫细胞，如 B 细胞、自然杀伤细胞等，共同参与机体的免疫防御。在多种免疫相关疾病中，CD2 及其相关信号通路的异常与疾病的发生发展紧密相连。在自身免疫性疾病，如类风湿关节炎中，T 细胞的过度活化是导致关节炎症和损伤的重要原因之一。研究发现，CD2 与 CD58 的异常结合会增强 T 细胞的活化程度，使其持续分泌大量炎症因子，如肿瘤坏死因子 -α（TNF-α）、白细胞介素 - 6（IL-6）等，加剧关节滑膜的炎症反应和骨质破坏。在某些血液系统恶性肿瘤，如 T 细胞白血病中，CD2 的异常表达和功能失调可能导致 T 细胞的恶性增殖和分化异常，逃避机体的免疫监视。
