p21（CDKN1A），也称为 WAF1、CIP1、SDI1，是 细胞周期蛋白依赖性激酶抑制剂（CKI） 家族的重要成员。p21通过结合并抑制 CDK1、CDK2和CDK4/6 的活性，在 细胞周期G1/S期转换 中发挥关键负调控作用，是细胞周期停滞和DNA损伤修复的核心调控因子。
p21是 p53肿瘤抑制通路 的重要下游靶基因，在DNA损伤、氧化应激和癌基因激活等条件下被诱导表达，介导 细胞周期停滞、细胞衰老和凋亡。p21的表达异常与多种 恶性肿瘤 密切相关，包括 结直肠癌、肺癌和乳腺癌 等。此外，p21在 干细胞维持、组织再生和免疫调节 中也发挥重要作用，已成为肿瘤治疗和衰老研究的重要靶点。
