二、产品描述
最初的小鼠IgG1抗体可与所有稀土的DOTA螯合剂结合，因此可用于捕获各种性质的探针分子。该抗体的原始IgG1版本曾被用于对患有KHJJ肿瘤的BALB/c小鼠进行放射免疫疗法的预定位。体内清除研究表明，与金属螯合物结合的2D12.5有74-96%在服用结合复合物 24小时后仍然存在。该抗体的一个工程化版本在重链的第54、55或56位（靠近蛋白质结合位点的一个位点）有一个半胱氨酸残基，可形成 (S)-2-（4-丙烯酰胺基苄基）-DOTA (AABD)的弱亲电金属复合物，这些复合物可结合并形成永久连接。重链G54C突变体有一小部分亲电配体，反应活性的差异表明亲电原子附近的取代基可能是永久结合的重要决定因素。
